I have not tried to use the CB dies to cut acetate, but it does cut through chipboard. My guess is that you can cut through acetate w/the CB dies.

The plates will definitely get the etching/die outline on it. Do not fret. It will takes lots and lots and lots of the CB use before the plates gets so etched that you will be unable to use it....
